我们已经证明滋阴清热中药对SLE基因和蛋白有明确调控作用,因狼疮活动指数的缺陷,严重干扰了调控效能判断和后续作用机制研究。细胞焦亡(Pyroptosis)是新近研究热点,焦亡时对邻近细胞产生强烈的促炎信号,引起明显细胞损害,其负反馈失调炎症进行性加重。据此我们推测焦亡可能与狼疮活动密切相关,但迄今未见报道,我们前期已证实焦亡负反馈相关基因atg16l1参与狼疮活动。本项目为研究滋阴清热中药干预效果,首次将焦亡引入狼疮活动评估领域,通过三种危险因素梯度浓度诱发经地塞米松预处理后的狼疮鼠,建立狼疮鼠轻度活动梯次模型,给予滋阴清热中药干预,采用基因沉默等技术,以足细胞为载体系统,观察细胞焦亡及负反馈信号通路在狼疮活动和器官损害中的作用,同时明确中药对焦亡及负反馈信号通路的影响。研究将可能建立新的狼疮活动评价指数及筛选活动标志物,发现狼疮活动新的发病机制,阐明中医药的效能及作用机制。

We have proved that Ziyinqingre Chinese drugs have the clear regulatory role on SLE gene and protein. as the activity index defect we could not judge its regulatory efficacy and difficult continue follow-up mechanism research. pyroptosis is recent research focus. when pyroptosis occur cell could produce pro-inflammatory signals to neighboring cells and cause significant cell damage. If its negative feedback disorders the inflammation increase progressively . Accordingly, we speculated that pyroptosis is closely related to lupus activity. But so far no any report We have confirmed that atg16l1 genes related to pyroptosis negative feedback have involved in lupus activity. For study the efficacy Ziyinqingre Chinese drugs in this project we first Introduce pyroptosis to the assessment lupus activity and build slight lupus activity gradient model by the method that inducing lupus mice lupus activity with three risk factors gradient concentration induced after dexamethasone pretreatment lupus mouse. giving Ziyinqingre Chinese drugs intervention and using technologies such as gene silencing to systematic observe pyroptosis and its negative feedback signaling pathway in lupus activity and organ damage development and define the effect of Ziyinqingre Chinese drugs on pyroptosis and its negative feedback signaling pathway The research will be to establish possibly the new lupus activity evaluation index and screen activity markers ,discovery the new pathogenesis of lupus activity ,clarify the efficacy and mechanism of Traditional Chinese Medicine
