前期研究发现艾灸能够良性调控肺纤维化TGF-β/IFN-γ信号通路,该通路与纤维化核心机制- - 上皮细胞-间质细胞转分化(EMT)密切相关。本课题基于EMT研究前沿,探讨艾灸"塞因塞用-通补兼施"治则调控肺纤维化EMT的分子机制。拟复制经典肺纤维化大鼠模型,动态观察造模及艾灸治疗过程中,肺组织EMT进程重要组分及关键调控因子(E-钙粘蛋白,MMP1,TIMP1,vimentin,α-SMA等)基因表达及蛋白分布规律,揭示艾灸拮抗肺纤维化之EMT关键靶点与网络调控模式。同时,分离不同时间窗口肺上皮细胞,原代培养并检测前述EMT关键分子含量与变化规律,探讨艾灸拮抗该病EMT的动态调控机制。本课题整合EMT理论成果及研究方法,揭示艾灸治疗肺纤维化之EMT调控规律,为中医药防治器官纤维化提供新思路,为确立艾灸防治肺纤维化临床方案提供新的科学依据,为提升肺纤维化中医药临床疗效奠定坚实基础。
英文摘要
Based on Traditional Chinese Medicine theory, the research will set up the project in PF- EMT- TGF-β/IFN-γ. TGF-β and IFN-γ receptor signaling pathways are positive significance on moxibustion resistance PF in pilot studies, so the research will explore the molecular mechanism of to reveal biological basis on moxibustion resistance PF. The research will select different development stages of the lung tissue in PF animal model, measure lung tissue in the body, organization purification epithelial cells and EMT chemotactic in vitro testing. The research will detect collagen-1mRNA, protein microarray, TIMP-1、TIMP-2 and RT-PCR, to screen the regulation network and key targets on moxibustion resistance PF in EMT level, and multiple perspectives analysis moxibustion resistance EMT molecular mechanism from TGF-beta and IFN-γ key pathway based on the information fusion method.
