血吸虫病流行广泛,危害严重。血吸虫雌雄合抱是雌虫发育成熟的前提,成熟雌虫所产的大量虫卵又是造成宿主主要病理损害和疾病再传播的原因。血吸虫雄虫特异表达的抱雌沟蛋白具有影响雌雄合抱的效应。本课题在已有基础上,利用酵母双杂交技术,对日本血吸虫抱雌沟蛋白相互作用分子进行了筛选,最终获得23个在酵母体系内与血吸虫抱雌沟蛋白相互作用的分子。利用免疫共沉淀(Co-IP)技术证明在哺乳动物细胞体系内抱雌沟蛋白与SjLGL,SjCol-Ⅴ间存在着相互作用,免疫组化技术也证实SjGALE,SjGAPDH-24,Sj34.9,SjActin-а-2,SjTWF与抱雌沟蛋白在空间结构上的存在相互作用性的可能性。利用基因芯片杂交、RNA干扰、Real-time PCR等技术研究发现抱雌沟蛋白及其相互作用分子对血吸虫生长发育、形态结构、虫卵孵化(减虫卵孵化率可高达85%)、雌虫产卵能力等有显著影响。对其及相互作用分子诱导的免疫机制分析发现可以诱导机体产生对寄生虫感染具有保护性的Th1型免疫反应。本研究对阐明雌雄合抱和血吸虫发育生殖的调控机制有重要理论价值,对开拓抗血吸虫生殖发育新途径有实践意义。

Schiatosomiasis is a globally important helminthic disease of humans and animals, and is the second most common parasitic disease after malaria, even though treatment has been available for two decades.Unlike other trematode parasites, schistosomes have evolved separate sexes or dioecy. The dioecious state created an opportunity for the sexes to play a role in schistosome evolution that has resulted in an interesting interplay between the sexes. Studies up through the 1990s focused on dissecting the role of the sexes in mate attraction, mate choice, mating behavior, female growth, female reproductive development, egg production, and other sex-evolved functions.The classical observation is the female schistosomes do not complete reproductive development unless a male worm is present. The male by an unknown mechanism transduces a signal that regulates female gene expression in a stage-, tissue-, and temporal-specific manner. The Gynecophoral Canal Protein (Gcp) of Schistosome is characterized to be a glycoprotein which exhibits surface expression in adult parasites especially in male worms in which expression is limited to the gynaecophoric canal , the site of direct interaction between the mating pair. Interestingly, the transcription profile of Gcp shows its peak expression in a time that coincides with worm pairing. This distribution pattern emphasizes the potential role of GCP in male–female interactions and the male-stimulated reproductive maturation of the female schistosome.In this study the yeast two—hybrid technique was used to screen the intracellular proteins interacting with GCP of schistosome japonicum,which may be involved in the female worms mature induced by male worms.In this report the SjGcp coding sequence was amplified by polymerase chain reaction method and was cloned into the bait plasmid pGBKT7 vector,which,after the confirmation by sequencing,was transformed into yeast AHl09 and the subsequent expression of bait plasmid was proved by Western—blot.The transformed yeast AHl09 was mated with yeast Y187(containing 18 days Schistosomula japonicum cDNA library plasmids pACT2)in medium.Diploid yeast was plated on synthetic dropout nutrient medium containing X- - gal for screening.Finally,after being reintroduced into yeast AHl09 and sequenced to verify the expression of ORF,23 proteins were defined is interacted with SjGCP in yeast system.The interaction between SjGCP and SjLGL,SjCol-were retested in mammalian cells by coimmunoprecipitation.The possibility of interaction between SjGCP with SjGALE,SjGAPDH-24,Sj34.9,SjActin-а-2,SjTWF were confirmed by indirectimmunolocalization For study the biology function of these interaction factors, the gene chip ,RNA interfere, Real-time PCR and so on techniques were used. The results showed these protein have important roles in the schistosome development, the parasite morphology, the eggs hatch(the effection on hatch rate high to 80%), the female worms reprodictive ability. The study on immune mechanism showed the Th1 type protective immunity to parasites were reduced.
