目 的:观察慢性鼻窦炎(CRS)鼻腔黏膜及黏液中嗜酸性粒细胞(EOS)表达情况,明确嗜酸性粒细胞主要碱性蛋白(MBP)在CRS鼻腔黏液中的表达; 探讨交链霉孢菌抗原对CRS患者外周血单核细胞来源的树突状细胞的免疫功能的影响。探讨真菌引起的异常的免疫反应导致CRS的可能性。材料与方法 : 1.CRS鼻腔黏液中MBP的表达及意义研究:取CRS鼻腔黏膜黏液及健康人鼻腔黏膜黏液,应用苏木素-伊红染色法观察黏膜及黏液中EOS表达,应用糖原染色法观察黏膜及黏液中真菌,并应用酶联免疫吸附法检测黏液MBP的表达。2.真菌对慢性鼻窦炎的树突状细胞的生物免疫学功能的影响研究:分别采集CRS患者和健康志愿者的外周血,分离外周血单核细胞进行培养,每例单核细胞分别在肿瘤坏死因子α(TNF-α)和真菌抗原共同刺激下或TNF-α单纯刺激下增殖成熟,应用间接免疫荧光流式细胞法检测每份DC的表型。结 果:1. CRS鼻腔黏液中MBP的表达高于对照组(p<0.01)。2. 真菌抗原刺激后CD86,CD80表达的变化与对照组有显著差异(p<0.05)。结 论 : CRS鼻腔黏液中MBP表达增加可能是CRS发生的原因。

Objective To observe eosinophils (EOS) located in mucosa and mucus in Chronic Rhinosinusitis(CRS), to determine whether migration of eosinophils and their activation by releasing eosinophil major basic protein (MBP) in the extramucosa fluid of CRS patients; To compare immunologic response of dendritic cell (DC) between CRS patients and non-CRS controls after in vitro stimulation with Alternaria. To find evidence hypothesis in the Pathophysiology of Chronic Rhinosinusitis. Methods 1. Expression and its significance of eosinophil major basic protein in chronic rhinosinusitis: Mucosa and mucus obtained from CRS paitents and health controls were stained by Heamatoxylin and eosin(H&E)for eosinophils count; by periodic acid-Schiff reaction(PAS) for fungal hypha .Thirty-nine nasal mucus obtained from CRS patients were used to detect the expression of MBP by Elisa assay. Ten nasal mucus samples from health people were used as control. 2. The effect of Alternaria on the bioimmunological function of dentritic cell in chronic rhinosinusitis: Mononuclear cells were isolated from patient's peripheral blood by density gradient centrifugation, and adhered cells were cultured in medium containing GM-CSF, after 5 days, TNF-α, and 1 more days, alternaria antigen were added in half of cells, after 1 more days, monocyte-drived DCs
