喜马拉雅紫茉莉作为特色的藏药资源,具有重要的药用开发价值。开发利用导致其野生种质资源的生存受到严重威胁。而对其保护一直未引起足够重视,对其进行的资源保存也主要出于经济目的,比较随机和盲目。此外,通过前期研究发现,其种质资源的形态变异丰富,一些重要性状描述不准确,分类鉴定还存在问题,需要明晰。鉴于此,本研究拟对西藏分布的喜马拉雅紫茉莉野生种质资源进行全面系统地调查,通过形态、ITS序列和cpDNA非编码区片段分析,明确其鉴定问题,为其科学研究和开发利用提供基础;利用SSR分子标记与形态学分析结合,研究其遗传多样性的水平和结构,旨在建立藏药材喜马拉雅紫茉莉的遗传背景资料,为其种质资源的有效保护和合理利用提供科学可靠的理论依据;同时,基于野生居群遗传多样性数据的筛选分析,确定核心种质最佳样本选择方案,为后期其核心种质的构建奠定基础。
英文摘要
Being special tibetan herbal medicine, Mirabilis himalaica has great medical development value.Its wild germplasm resources are in danger because of exploition, but the protection dose not receive enough attention. In addition, the conservation of its germplasm resources are blindfold and for economic benefits only at present.Through prophase research, We discover that there is some confusion in its classification and identification, and some primary identification characteristics are not accurate. In view of this, we will investigate the wild germplasm resources of M. himalaica in tibet systematically and make its identification definite through morphology、ITS sequence、noncoding cpDNA region analyses. The study will provide basic imformation for scientific research and exploition on M. himalaica . Meanwhile, the genetic diversity of wild germplasm resources will be studied by means of morphological analyses and SSR molecular markers,which will establish the genetic background of M. himalaica. Furthermore, Based on the analysis of genetic diversity, we will offer scientific advices on its conservation and exploitation effectively. At the same time, The strategy for conservation sampling of core collection will be determined by screening and analyzing genetic diversity data of wild populations accordingly, which will lay a foundation for the construction of core collection.
